Preparation, characterization, and ex <i>vivo</i> mucoadhesive studies of triamcinolone acetonide-impregnated Chitosan/PLGA nanocomposite buccal film for oral mucosa ulcer

In spite of the numerous approaches have been taken up to prevent and treat oral mucosa ulcer, the formulations existing in the market (mouthwash, paste, ointment, gel) are not fully effective. Triamicilone acetonide (TAA) is a synthetic class IV corticosteroid having anti-inflammatory properties and commonly used to relieve the discomfort of a mouth sore. The use of poly (lactic-co-glycolic acid) based nanoparticles (PLGA NPs) as drug delivery carriers is highly recognized due to their biocompatibility. The purpose of this study was to conduct a prospective evaluation of nanocomposite chitosan mucoadhesive buccal films that contain TAA-loaded PLGA nanoparticle as a drug delivery system for the treatment of oral mucosa ulcer. TAA-loaded PLGA nanoparticles were prepared by single emulsion techniques, with different ratios of nanoparticles embedded with chitosan in order to obtain nanocomposite mucoadhesive buccal films. The average particle size of the formulation was 184 nm. In Fourier transform infrared spectroscopy (FTIR) analysis, there was no significant interaction between the medication and polymer utilized in this study, and the scanning electron microscopy (SEM) image showed a continuous and compact section structure. The release rate of nanocomposite mucoadhesive buccal film formulation F2 and F6 was 96.01% and 95.68%, respectively, among the developed formulations.
Ex-vivo
residence time confirmed that all the films were perfect and had good swelling and drug release properties. Based on these results, this study concluded that chitosan buccal films containing TAA-loaded PLGA nanoparticles as promising approach for the treatment of oral mucosa ulcer.
